ABOUT THE COURSE: Transgenesis is the process of introducing changes into the genome of an organism to create a transgenic organism. Transgenic animals are valuable for fundamental studies of biological phenomena and have significant relevance in the development of highly specific therapeutics and biologics. The technology for creating transgenic mice has evolved rapidly, progressing from traditional recombination methods to modern CRISPR-Cas9-mediated techniques.In this course, I will discuss the principles and methods of animal transgenesis, with a particular focus on miceINTENDED AUDIENCE: Final year BS/BSc students studying subjects related to animal/human, BVSc, MBBS, MS/MSc students studying subjects related to animal/human, MVSc, and PhD students.PREREQUISITES: For doing the course is that students/candidates should have good basic knowledge of "Molecular Biology", "Biochemistry", "Genetic Manipulation/Genetic engineering", and "Immunology".INDUSTRY SUPPORT: Any biotechnology industry

Syllabus

Week 1: Introduction of transgenesis; Brief History of transgenesis; why we need transgenesis? Model organisms for transgenesis and application of various transgenic animals in basic discoveries and translational research in development of clinical general/precision therapeutics; Practical Application of 3Rs (Replace, Reduce, Refine) in Mouse
Week 2:General principles of transgenesis in Mouse; Tools and Technique for creation of transgenic mice; creation and methods for gene overexpressing mice; Methods for animal/mammalian cell culturing;
Week 3:Tools and Technique for creation of transgenic mice using homologous recombination method. Creation of Gene-deficient/knockout mice using homologous recombination method. Creation of conditional or cell-type specific- or tissue type-specific gene-deficient mice.
